B-590.45.009 [2020-10] Carotenoids and Total Carotene – iCheck Method/HPLC

Numerous beverages, such as multivitamin juice and sports drinks, are enriched with vitamins. These beverages may not be listed on a label as "enriched" or "fortified" with a particular vitamin supplement unless the vitamin is present in "significant amounts", which are defined in Annex XIII to Regulation (EU) No 1169/2011. For beverages, this amount is set at a minimum of 7.5 % of the NRV (nutrient reference values) per 100 ml or 15 % of the NRV per portion for individual portions.

Quantities recommended for the daily dietary intake of vitamins:

vitamin B₁ (thiamine)	1.1	mg

vitamin B₆ (pyridoxine)	1.4	mg
pantothenic acid	6	mg
folic acid	200	µg
niacin	16	mg
vitamin B₁₂	2.5	µg
ascorbic acid	80	mg
vitamin E	12	mg
biotin	50	µg
vitamin A*	800	µg

* 1 µg vitamin A corresponds to 6 µg all-trans-β-carotene or 12 µg of other provitamin-A carotenoids.

Approx. 50 carotenoids exhibit a provitamin-A effect. Within the confines of a normal diet, however, only α-carotene, β-carotene and β-cryptoxanthin are of any significance.

Determination of the concentration of carotenoids and provitamin A

Scope of Application

This method is suitable for NAB and juice.

Principle

The carotenoids are extracted with a solvent. The total carotenoid content in this solution is determined photometrically. After dissolving in ethanol, the content of β-carotene and α-carotene is determined using HPLC through separation on a C30 phase. The vitamin A content is then calculated.
